Guanggancao Ding is a natural isoflavone with estrogen receptor agonists and significant anti-tumor activity. In addition, Guanggancao Ding has a regulatory effect on the PI3K/AKT/mTOR pathway, but its exact target is still unclear. Lin Hongyan, a teacher from Changzhou University, further identified its targeting PI3K by evaluating the antitumor activity of glabridin on breast cancer and prostate cancer cells. It has been found that glabridin can significantly inhibit the cell viability of human breast cancer and prostate cancer cell lines. It induces caspase cascade activation and cell apoptosis by reducing mitochondrial transmembrane potential and increasing intracellular reactive oxygen species (ROS). Elucidating the targeting effect of glycyrrhizin on PI3K may lay a theoretical foundation for the structural derivatization of glycyrrhizin, and is expected to greatly promote its application and development in the field of cancer treatment.
