Question 10: Why is there a batch number difference in the cell culture effect of serum, and how can batch number differences be avoided as much as possible?
Because serum comes from animals and has around 1000 components, the composition and content of each batch are uncertain, and there may be differences between batches. We conduct the following three important performance tests on each batch of serum products: continuous observation of cell morphology for three generations, 72 hour cell expansion factor, and cell viability detection.
Question 11: How can different brands and product numbers of serum be replaced?
When changing serum of different brands or product numbers, some steps need to be taken to ensure the continuity of cell culture and the growth status of cells. 1. Pre testing: Before a comprehensive replacement, conduct small-scale testing on the new serum to evaluate its impact on the growth and characteristics of the cell type you are cultivating. 2. Gradual transition: If possible, do not suddenly replace the serum completely. Cells can adapt by gradually increasing the proportion of new serum. For example, at the beginning, the new and old serum can be mixed in a 1:1 ratio, and then the proportion of new serum can be gradually increased. 3. Observing cellular responses: During the replacement process, closely monitor the morphology, growth rate, and any possible abnormal manifestations of the cells. 4. Recording and comparison: Record the observation results during the replacement process and compare them with the data obtained when using old serum. 5. Maintain consistency in other conditions: While changing the serum, try to keep the culture medium and other culture conditions unchanged to reduce the impact of other variables on the cells. 6. Consider cell types: Different cells have different adaptations to serum, and some cells may be more sensitive to serum replacement. 7. Pay attention to serum characteristics: Understand the characteristics of different sera, including their sources, endotoxin levels, growth factor content, etc., and choose the most suitable serum for cell types. 8. Follow supplier guidelines: Different serum suppliers may provide specific replacement guidelines or recommendations, and following these recommendations can improve the success rate of replacement.
