       Oxaliplatin (OXL) is a first-line drug for the treatment of colon cancer, with significant therapeutic effects. Gut toxicity is a common side effect of oxidants, with unclear pathogenesis and a lack of effective treatment strategies. Polygonatum sibiricum glycoside (PD) has anti-inflammatory and antioxidant effects and is a potential drug for treating intestinal diseases, but its poor water solubility limits its application. The team led by Professor Guo Chuanlong from Qingdao University of Science and Technology used polyvinylpyrrolidone (PVP) as a carrier to prepare PD (PVP-PD) nanoparticles with a particle size of 92.42 nanometers and sustainable release properties. The in vitro experimental results showed that PVP-PD protected NMC460 cells from oxidative damage, mitochondrial membrane potential (MMP) disruption, and accumulation of reactive oxygen species (ROS). The in vivo results demonstrate that PVP-PD has a protective effect against OXL induced intestinal toxicity, such as reducing oxidative induced weight loss and shortening of colon length. The in vivo and in vitro mechanisms indicate that OXL can induce DNA damage, activate the cGAS/STING pathway, and further induce the expression of inflammatory factors such as IL-1 β and TNF - α. PVP-PD alleviates the aforementioned changes caused by OXL by inhibiting the DNA damage cGAS/STING pathway. The research results indicate that PVP-PD provides a possible strategy for treating OXL induced intestinal toxicity.
