Ghrelin regulates various physiological activities. However, the effect of this hormone on milk fat synthesis is still unclear. The team led by Professor Wang Jiaxin from Jilin University studied the effects of acylated ghrelin (AG) on the expression of growth hormone secretagogue receptor 1a (GHSR1a) and th induced POK (ThPOK) in primary bovine mammary epithelial cells (BMECs), and explored the impact of AG on milk fat synthesis. The results showed that AG significantly increased the relative content of triglycerides in BMECs through GHSR1a, from 260.83 ± 9.87 to 541.67 ± 8.38. ThPOK, as a key regulatory target downstream of AG, promotes milk fat synthesis in BMECs by activating the PI3K and mTOR signaling pathways. In addition, AG regulates ThPOK by increasing EP300 activity, thereby promoting ThPOK acetylation to protect it from proteasomal degradation. In summary, AG increases ThPOK acetylation and stabilizes ThPOK through GHSR1a, thereby activating the PI3K/mTOR signaling pathway and ultimately promoting milk fat synthesis in BMECs.
