Ⅰ.Article Introduction
Tripterine (TP) is a pentacyclic triterpene extracted from Tripterygium wilfordii, which has beneficial effects on obesity, diabetes and other diseases. However, the effect of TP on beta cell lipotoxicity has not been fully explored. Here, the team led by Professor Wang Zhiyong from Zunyi Medical University discovered that TP regulates β - cell lipotoxicity in a concentration dependent and bidirectional manner. At low concentrations, TP may protect MIN6 cells from palmitate (PA) - induced lipotoxicity. At high concentrations, TP significantly promotes beta cell lipotoxicity, further enhancing PA induced cell apoptosis. In addition, low concentrations of TP inhibit the increase in reactive oxygen species (ROS) levels induced by PA, and its protective effect is eliminated by the ROS inducer tert butyl hydroperoxide. On the contrary, high concentrations of TP significantly intensified PA triggered ROS generation, and its enhanced cytotoxicity was partially reversed by ROS inhibitor N-acetyl-L-cysteine. Therefore, TP plays a dual role in β - cell lipotoxicity, suggesting that caution should be taken when it is used in the treatment of obesity and diabetes.
